The Common Technical Document, harmonised through ICH, gives regulators in the United States, European Union, Japan and an expanding set of other markets a single dossier architecture. ANDA submissions and generic approval pathways

An Abbreviated New Drug Application is the pathway through which a generic medicine is approved in the United States. The application does not repeat the safety and efficacy programme of the innovator; instead it establishes that the proposed product is pharmaceutically equivalent and bioequivalent to the reference listed drug, and that it is manufactured under adequate quality controls.

The critical components are therefore the bioequivalence study or biowaiver justification, a complete Module 3 quality section demonstrating a controlled and reproducible manufacturing process, appropriate patent certification, and labelling that appropriately matches the reference product.

Approval timelines are dominated by review cycles rather than by initial preparation. Applications that receive complete response letters usually do so for quality section deficiencies, facility inspection outcomes or bioequivalence design issues — which is why cross-functional review before filing consistently outperforms speed of filing.

Drug Master Files and supplier documentation

A Drug Master File is a confidential submission containing detailed information about facilities, processes or articles used in the manufacture, processing, packaging and storage of a drug — most commonly the active pharmaceutical ingredient. It allows an API manufacturer to share proprietary detail with the regulator while granting only authorised access to the dossier holder through a letter of authorisation.

The DMF is conventionally divided into an open part, available to the applicant, and a restricted part, available only to the agency. Regulatory professionals working on finished products must be able to read the open part critically, reconcile it against the finished product specifications, and identify gaps that will predictably generate deficiencies.

Post-approval change and lifecycle management

Approval is the beginning of the regulatory workload rather than the end. Sites change, suppliers change, equipment is replaced, specifications are tightened, shelf life is extended, and each of these must be evaluated for regulatory impact and reported through the correct mechanism in each market where the product is registered.

The classification decision — whether a change is a major variation, a minor variation requiring notification, or a change reportable in an annual report — is the highest-value judgement a regulatory professional makes.
